From Orthosiphon stamineus Benth, seven pimarane diterpenes (1–7) have been isolated. By isolation by assay-guided. Both isolates had a 2-deoxy-2-((7-nitro-2,1,3-benzoxadiazole-4-yl)amino)-d-glucose absorption effect at 5 and 10 μM concentrations in 3T3-L1 adipocytes. Most of them demonstrated potent protein tyrosine phosphatase 1B inhibition with IC50 values ranging from 0.33 to 9.84 μM. In the kinetic analysis, all forms of inhibition were exposed to the forces studied, including mixed-competitive (1), non-competitive (3 and 5), competitive (6) and non-competitive (6) powers (7). The findings have indicated that O. Stamineus and its pimarane diterpenes could exert the hypoglycemic effect via the pathway of insulin signaling targeting protein tyrosine phosphatase 1B (PTP1B) activity inhibition. The findings indicate that these active constituents of O. Stamineus is a possible natural product for the production of antihypoglycemic agents.
